(a) Human ileum-derived and (b) human colon-derived epithelial monolayers cultured on transwell inserts under noninflammatory conditions. Both form confluent, polarized epithelial layers with intact morphology, demonstrating the successful establishment of primary human intestinal monolayers from donor tissue.
(a) Transepithelial electrical resistance (TEER) measurements showing a notable reduction in TEER in cytokine-treated (IFNγ, IL-1β, TNFα) inflamed monolayers compared to untreated controls (p < 0.001). (b) Fluorescent dextran permeability assays using lucifer yellow, 10 kDa, and 70 kDa dextrans demonstrate significantly increased permeability for lucifer yellow and 10 kDa dextrans in inflamed monolayers, with minimal change for 70 kDa dextran, indicating cytokine-induced compromise of epithelial barrier integrity in human colon monolayers.
